人谷胱甘肽還原酶(GR)ELISA檢測試劑盒試驗以靈敏度較高、特異性較好的特點在臨床上得到了廣泛的應用,但操作中的各個環(huán)節(jié)對試驗的檢測效果影響較大,如不注意,有可能導致顯色不全、花板等結果。我將操作中各個環(huán)節(jié)常出現(xiàn)問題的原因及解決辦法總結于下,以期給同行帶來一些啟發(fā),提高試驗質量。
下面分析Elisa試驗操作中可能影響結果的原因,并給出相應的解決辦法
1 、選擇試劑
選擇質量優(yōu)良的檢測試劑,嚴格按照試劑說明書進行操作,操作前將試劑在室溫下平衡30-60分鐘。
2 、加樣
可能原因: 1)血清或血漿標本分離不好即進行加樣; 2)手工操作中,加樣板過多造成加樣后放入孵箱前等待時間過長(特別是室內溫度較高時); 3)加完標本再加酶試劑時酶濺出孔外。
解決辦法: 1) 標本為血清:將血液先自然存放1-2小時后,再用3000rmp離心15分鐘;標本為血漿:必須使用含抗凝劑的血液標本收集管,采血后必須立即顛倒采血管混合5-10次,放置一段時間后,3000rpm離心15分鐘;若在幾天內檢測,可放在2-8℃冰箱中,若要貯存,則置于-20℃的低溫冰箱內。 2) 加樣后及時放入孵箱。 3) 加酶試劑后用吸水紙在酶標板表面輕拭吸干。 4) 如果采用AT或其他全自動加樣,選擇FAME或其他后處理儀器加酶試劑。 5) 標本較多時,請分批操作。
3 、孵育
可能原因: 1) 孵育時未貼封片或加蓋,使標本或稀釋液蒸發(fā),吸附于孔壁,難于清洗*; 2) 孵育時間人為延長,導致非特異性結合緊附于反應孔周圍,難以清洗*。
解決辦法: 1) 貼封片或加蓋; 2) 按說明步驟嚴格控制操作時間。
4 、洗板
可能原因: 1) 采用手工洗板,孔與孔之間液體交叉。 2) 采用半自動洗板機洗板時,洗液量不足,導致洗板不*;洗板針堵塞,抽吸不*;洗板不暢,導致洗板效果差。 3) 反應板過多造成洗板等待時間長。
解決辦法: 1) 保證洗液注滿各孔,洗板針暢通,洗完板后在吸水紙(選擇干凈、無或少塵的吸水材料)上輕輕拍干; 2) 合理安排,或多用幾臺洗板機。
5 、顯色
可能原因: 1) 顯色劑配制后放置時間過長或使用過期顯色劑; 2) 加顯色劑時濺出孔外造成液體回流。
解決辦法: 1) 顯色劑盡量在臨用前配制,堅持不用過期顯色劑,肉眼可見淺藍色的TMB顯色劑不用; 2) 加樣時保持顯色劑不外流; 3) A、B液應避免接觸金屬器械。
6 、終止
可能原因如加終止液時產(chǎn)生較多氣泡,導致假陽性增加。所以在加終止液時應避免產(chǎn)生氣泡。
7 、讀板
如讀板時板底不清潔等。應保證酶標板清潔。
所以在整個操作過程中保證酶標板不接觸次氯酸; 盡可能實現(xiàn)ELISA檢測標準自動化,有效提高檢測質量。
